Question about weight loss

Ok, I have a quick question which I've been pondering for a while so I'll just go ahead and ask. My question is, is it possible to undo a whole week's worth of losing weight in one day? For instance, if I'm counting calories all week, going to the gym 4 times doing a mixture of cardio and weights etc, could I consume 4000 calories in one day and undo all of the work I've done throughout the week?

The reason I ask is that I unfortunately am partial to a bit of a drink, so some friday nights I will go out and consume a fair amount of calories in beer. I know I shouldn't be drinking and I have cut down, but I was just curious to know if I could be undoing a whole week's worth of work in one hit? Does weight loss work that way?
